lucagon-like peptide-2 (GLP-2) is a potent intestinotrophic/satiety hormone that acts through its G protein-coupled receptors (GLP1 and GLP2R). Tissue-specific posttranslational processing of proglucagon leads to GLP-2 and GLP-1 secretion from the intestine and glucagon secretion from the pancreas. Peptide hormones of the secretin-glucagon family are mainly expressed in endocrine cells of the pancreas, gastrointestinal epithelium and in specialized neurons in the brain (1). These peptide hormones subserve multiple biological functions, including regulation of growth, nutrient transit within the gut, digestion, energy absorption, and energy assimilation. The actions of these peptides are mediated by distinct members of the glucagon receptor super-family, all G protein-coupled receptors (GPCRs). GLP-2 also reduces gut permeability, bacterial translocation and pro-inflammatory cytokine expression under conditions of intestinal inflammation and injury. Glucagon, glucagon-like peptide-1 (GLP1), GLP2, glucose-dependent insulinotropic peptide, growth hormone-releasing hormone and secretin are structurally related peptides that exert their actions through members of related G protein-coupled receptor class 2 family (2). These receptors exhibit tissuespecific expression and significant structural homology associated with GPCRs (7 transmembrane domains) and share similar structural and functional properties with respect to ligand binding and signal transduction. GLP-2R expression is detected in multiple extrahypothalamic regions of the mouse and rat CNS, including cell groups in the cerebellum, medulla, amygdala, hippocampus, dentate gyrus, pons, cerebral cortex, and pituitary. The trophic actions of GLP-2 are specific for the intestine and are mediated by stimulation of crypt cell proliferation and reduced apoptosis in mucosal epithelial cells (2). Central infusion of GLP2 led to suppression of food intake (3). The effects of GLP-2 are mediated by GProtein coupled GLP2-receptor, that is localized to the neuronal and endocrine cells of intestinal mucosa, but not in enterocytes, and hypothalamus which suggests that its actions are mediated indirectly via a secondary signaling mechanism. Glucagon Like peptide 2 receptor (PGL2R) is G-protein coupled receptor containing 7 transmembrane domains (TMD) with 553 amino acids (66.7 kDa). The GLP2 receptor exhibit unique patterns of tissue-specific expression, considerable amino acid sequence identity, and share similar structural and functional properties with respect to other glucagons like peptide receptors. The GLP2R-selective antibodies were generated against circular peptide unique to GLP2 receptor protein. The GLP2R antibodies have been fully characterized for cross reactivity with GLP1R and for other GPCRs.
